Depending upon the html of the page you may or may not be able to determine if there is a watermark.
If the site you are scanning has img tags that consistently differentiates a watermarked image and a non-watermarked image you could either use a Set Named Tag Step or a Test Tag step using a Tag Pattern. However, if the img tag is generic, you would not be able to use Kapow to determine if an image contained a watermark.
For example, the tag below contains both watermark and wm.jpg. Your Set Named Tag or Test Tag would be able to find/extract/test the tag.

This next tag is an example where you would not be able to determine if an img contained a watermark:
